    Therapeutic Category

Sildenafil: Phosphodiesterase type 5 (PDE 5) inhibitors

    How Sildenafil works

Sildenafil works by blocking the effects of certain enzymes (PDE5) within the body which limits blood flow to the lungs and penis. As a result, it causes widening of blood vessels (arteries) supplying these organs, increases blood flow and oxygen supply and improves their functions.

    Abstract

Treatments are required to the treatment of erectile dysfunction in men. They are required to be effective, with no side-effects, which is essential in the clinical setting. The first example of an effective drug is the selective inhibitor of phosphodiesterase type 5 (PDE5) from the penile area. A single dose of 10 mg of tadalafil has been tested in an animal model of erectile dysfunction. Tadalafil and sildenafil, which are similar in structure, were able to reverse the effect of PDE5 inhibitors on the erection in this animal model. The clinical studies performed in healthy men with no history of cardiovascular disease and no erectile dysfunction were not enough to determine the effect of tadalafil on erectile dysfunction, and it is not known whether sildenafil affects the erection of the penis. In addition, a positive effect of tadalafil on erectile dysfunction has not been demonstrated. In this study, we evaluated the effect of tadalafil on the penile erection in men with erectile dysfunction. We used the penile erection test, which was used in previous studies to test the effect of tadalafil on the erectile dysfunction of patients. After the test, we observed the effects of tadalafil and sildenafil in the penile erection in men with erectile dysfunction, which was confirmed by the penile erection test. After the test, the erectile dysfunction patients were again evaluated, and a negative effect was observed in the penile erection test. Our study shows that the effects of tadalafil and sildenafil on the erectile dysfunction of patients are similar to those of healthy men. Our results demonstrate that the effects of tadalafil and sildenafil on erectile dysfunction are similar to those of patients with a history of cardiovascular disease. These findings suggest that the use of tadalafil is safe and effective in the treatment of erectile dysfunction, and it is recommended to start treatment with the tadalafil in men.

Treatment of erectile dysfunction in men with pulmonary hypertension and a positive effect of tadalafil is an effective treatment with a good safety profile in the clinical setting. However, the safety of tadalafil was not assessed in clinical trials. We evaluated the effects of tadalafil on the penile erection of men with erectile dysfunction. We performed the test in healthy male volunteers, and we evaluated the effects of tadalafil on the penile erection of patients with a positive effect of tadalafil.

Note: Other medications can affect how Sildenafil is processed in your body, potentially impacting its effectiveness. These include azole antifungals (such as itraconazole or ketoconazole), macrolide antibiotics (such as clarithromycin or erythromycin), HIV protease inhibitors (such as saquinavir), mifepristone, rifampin, and ritonavir.

Combining Sildenafil with other medications for erectile dysfunction (ED) or pulmonary hypertension, such as tadalafil or vardenafil, is harmful.
